Botanical Profile

Aesculus hippocastanum is a large deciduous tree with palmately compound leaves, conspicuous upright flower clusters and glossy brown seeds. The seeds are particularly known for their saponin complex, aescin.

Aesculus hippocastanum L. is native to south-eastern Europe and is now widely planted in parks, avenues and cultivated landscapes. The leaves are large, opposite and palmately compound. In spring, upright, candle-like inflorescences appear, bearing white flowers with yellowish or reddish markings. The fruits are green, usually spiny capsules containing large, glossy brown seeds. These seeds are commonly called chestnuts but are botanically unrelated to edible sweet chestnuts. As a botanical raw material, the seeds are the principal focus; bark, leaves and flowers have different chemical profiles.

Phytochemistry

Horse chestnut seeds contain a complex mixture of triterpenoid saponins known as aescin or escin. They also contain flavonoids, coumarins, tannins, starch, proteins and other accompanying constituents. Bark and leaves contain, among other compounds, coumarin derivatives such as aesculin and aesculetin as well as flavonoids and tannins. The saponin-rich seed fraction must be clearly distinguished from raw seeds and other plant parts. Processing, extraction, purification and standardisation have a major influence on composition and safety.

Traditional Uses

Horse chestnut has been widely planted in Europe since the early modern period as an ornamental, shade and avenue tree. The seeds were traditionally used as a washing material and, after special processing, in some cases as animal feed, as well as in various folk contexts. Seed extracts became particularly important in European herbal-medicine practice. The glossy seeds are culturally familiar but must not be confused with edible sweet chestnuts. Historical information relates to specific preparations and does not establish the safety of raw seeds.

Current Research

Research focuses on standardised seed extracts, aescin fractions, vascular models, inflammation models, permeability studies and clinical studies of defined extracts. Analytical work centres on aescin content, saponin patterns, coumarin derivatives, purity and extract standardisation. Findings obtained with purified or standardised extracts cannot be transferred to raw seeds, leaves or bark. Defined extract types, dosage, accompanying constituents and contaminant control are essential to quality and safety.
